Acute oral toxicity of hydroalcoholic extract of Spondias mombin L. (jobo)

ABSTRACT

Introduction: Spondias mombin, known as jobo or plum, is a tree of wide tropical distribution which is common in the vicinity of rivers. It is traditionally used as an additive or food dyes, in ice cream making, beverages and for its medicinal properties as: antiseptic, antimicrobial, antioxidant, anticholesterolemic, antiepileptic.
Objective: To evaluate the toxicity of the hydroalcoholic extract of Spondias mombin L. leaves by class method (CTA).
Methods: Leaves of Spondias mombin were collected in El Caney, Santiago de Cuba. The extract was obtained by maceration and was used as alcohol-water solvents (70:30 %). It was concentrated with rotary evaporetor at a temperature of 40ºC up to 50 mL. International standards Guide 423 of the Organisation for Economic Co-operation and Development of the European Economic Community and as described in the Toximed Manual of Operational Working Procedures were followed. The dose tested was 2000 mg/kg of body weight, and a single dose was administered in 24 h. The trial lasted 14 days and the variable to be measured was percentage of survival and weight gain. In addition, clinical observation of signs and symptoms was performed. At the end, the animals were slaughtered by narcosis with ketamine-50 mg and necropsy was performed.
Results: 100 % survival was recorded, with normal animal behavior and normal postural reflex. The first seven days the weight increased although it was not significant; which did happen at 14 days. There were no pathological alterations attributable to the effect of the extract on systems, organs and tissues, at the dose used.
Conclusions: The toxicity study of the extract of Spondias mombin L. showed that a single dose is framed as unclassified according to the Shelede scale, under the experimental conditions used. It also established that the toxic dose of the extract is greater than 2 000 mg/kg in a single-dose study.
