Polymorphism -308G>A of the gene coding for tumor necrosis factor alpha, periodontitis and acute coronary syndrome

ABSTRACT
Introduction: Periodontitis and cardiovascular diseases of ischemic origin have become a serious public health problem worldwide. These diseases have a multifactorial etiology with an important genetic component.Objective: To determine the behavior of the polymorphism of the tumor necrosis factor alpha -308G>A (rs1800629) with periodontitis and acute coronary syndrome.
Method: An analytical study of cases (patients with acute coronary syndrome) and controls was performed in the period from January 2019 to October 2020. The periodontal condition of the subjects was assessed using the Russell criteria. The -308G>A polymorphism was determined by polymerase chain reaction (PCR)/restriction fragment length polymorphisms (RFLP) following standardized procedures. In order to process the primary data, descriptive statistical techniques were used. In addittion, for the analysis of the possible relationship between genotype and the occurrence of periodontitis, the Cramer's V correlation coefficient was employed.
Results: Seventy-one percent of the subjects included in the study had a homozygous genotype for the wild-type "G" allele. Only 4.3% of patients with acute coronary syndrome were homozygous for the alternative "A" allele; they also presented periodontitis. Out of the homozygous patients for the alternative allele, 66.7% belonged to the groups with acute coronary syndrome.
Conclusions: The functional polymorphism -308G>A in the tumor necrosis factor alpha gene, may be a risk factor for periodontitis and does not modify genetic susceptibility for acute coronary syndrome. No statistically significant association was found between this syndrome and the presence or not of periodontitis.
