Frequency of sister chromotid exchange in a population free of exposure to clastogenic agents.

ABSTRACT

Introduction. Sister chromatid exchange (SCE) is a normal cellular event, with a relatively constant basal frequency of spontaneous SCE by metaphase, which is variable between cells from different tissues. Individual and technical factors can produce variation in the basal frequency of SCE. Exposure to clastogens can produce an increase in SCE. In this study we describe SCE frequency in a population free of exposure to known clastogens. Material and methods. We included 50 individuals of both genders, from 10 to 59 years old. From each individual two blood cultures, additioned with bromodeoxiurideine (BrdU) at 0 and 24 h, were carried out to obtain metaphases with differential chromatid stain. From each culture, 25 metaphases were analized to obtain the x of SCE/cell/individual. Results. The x of SCE/cell/individual found in 0 h and 24 h cultures was 4.21±1.20 and 3.91±0.94 respectively, with a range and SD from 2.12 ±1.12 to 7.4 ±3.31. In 24 h cultures the range and SD found was from 2.12 ±1.3 to 7.08 ±2.75. The difference between both cultures was not significant (p ‹0.10). The age gruop with the highest number of SCE was from 20 to 29 years old, and no significant differences were found between each ten year age group. Weither were significant differences found between genders. Discussion. The results found in these individuals were similiar to those found in other populations, with the exception of the decade with the highest SCE. These results allow us to feel confident in the use of the SCE analysis.
