Early tolerance versus Shwartzman reaction in a mouse model of lung injury using Klebsiella pneumoniae lipopolysaccharide

ABSTRACT

A previous administration of a low dose of lipopolysacharide (LPS) induces a state of early tolerance against a further LPS inoculation enough to provoke shock and death. However, when a sublethal amount of LPS is systemically administered, after another small amount of LPS previously inoculated in the skin or other organ like the lung, a local Shwartzman reaction could occur. The objective of this study was to evaluate if a small dose of E. coli LPS administered intraperitoneally (IP), before or after a transthoracic (TT) challenge with K. pneumoniae LPS in mice, induces a benefitial or an adverse effect on the lung inflammatory response. Comparisons, employing lungwet weight as an indicator of inflammatory reaction and histopathological observations, were made. Results showed that either a previous or posterior LPS IP small dose is more phlogistic than TT induced lung lesion alone. However, only the posterior small dose of LPS IP provokes a significant aggravating effect over the previous induced TT lung lesion. This effect was considered as a Shwartzman reaction.
