Evaluation of an immunoenzymatic assay on nitrocellulose paper for serodiagnosis of human and bovine brucellosis

ABSTRACT

An immunoenzymatic assay was developed on nitrocellulose paper for diagnosing human and bovine brucellosis, using lipopolysaccharide of Brucella melitenis 16M as antigen. An indirect-ELISA was the gold-standard for calculating the epidemiological sensitivity and specificity and positive and negative predictive values; the results between both tests were analyzed with the kappa test with a confidence level of 95%. The results for the human serum samples were 64.86%, 71.23%, 53.33% y 80%, for clinical sensitivity and specificity and positive and negative predictive values, respectively; there were low agreement between both tests (kappa = 0.343; p ‹ 0.05). The results for bovine serum samples were 29.17%, 87.78%, 38.89% and 82.23%, respectively; showing inadequate agreement between the tests (kappa = 0.187; p ‹ 0.05). Additional surveys are required for improving the immunological and epidemiological features of this immunoenzymatic assay, before it can be considered as an alternative to the diagnosis of brucellosis.
