Alveolar bone osteoclasts and intercellular adhesion molecule 1 (icam-1) On periodontal ligament cells under mechanical stress in rats with diclofenac or nimesulid treatment

ABSTRACT

Nimesulide is a weak inhibitor of prostaglandines synthesis, is capable of disminish considerably the population of osteoclasts in a rat´s alveolar bone. The expression of adhesion molecules ICAM-1 are important for precursor osteoclast´s cells migrate, towards the sites of resorption. Diclofenac inhibits the expression of ICAM-1. In this study we use a model or orthodontical mechanical stress over alveolar bone in order to compare the effect of both NSAIDs on the population of osteoclasts and in the expression of ICAM-1., fifteen rats were given diclofenac and fifteen other rats were given nimesulide. The animals were sacrificed in group of 5 at 24, 48 and 72 hours. Five rats were used like basal control. Maxilar histological slides were made and were dyed with HE and with TRAP. They were also given an inmunohisochemical treatment with monoclonal antibodies anti- ICAM-1. Results: between diclofenac and nimesulide animals there was no significative difference found in the number of osteoclasts (p›.001), the difference between both experimental group and the control group was significative (p‹0.001). In relation to the expression of ICAM-1, the witnesses submitted to mechanical stress, the group treated with diclofenac the ICAM-1 expression decreased at 12, 24 and 48 hours, and the difference between this group and the control and the animals treated with nimesulide was significative (p=0.001).
