Effect of lymphocyte polyplidy / aneuploidy on fertility of holsein cows in the state of Zacatecas, Mexico.

ABSTRACT

In order to determine if polyploidy or aneuploidy in lymphocytes and parity had an effect on Holstein cows’ fertility, 107 Holstein cows from several dairy farms in the Mexican state of Zacatecas with ranging ages from three to ten years were analyzed for their chromosomal makeup and tested its association with the number of times a cow returns to service and parity. 7.0 ml blood samples were collected from the cows’ jugular vein in heparinized Vacutainer vials and taken to the Cytogenetics Laboratory at the University of Zacatecas Veterinary School to perform a standard karyotyping technique and a G chromosome banding assay. As result 26.8% (28) of cows showed normal karyotypes, whereas on 73.2% (79) varying degrees of polyploidy or aneuploidy were observed on these cows’ lymphocytes metaphases, yielding from 1.0% to 16.0% defective karyotypes. Poisson regression showed significance (p ‹ 0.05) on associations between polyploidy and the number of times a cow returned to service; and between parity and the number of times a cow returned to service nor interaction neither association were found (p › 0.05) between polyploidy or aneuploidy and parity. It can be predicted that cows having 8.0% or greater percentages of polyploidy or aneuploidy will have their return to service rates significantly increased. It can be speculated that, in this study, polyploidy or aneuploid oocytes, which were ovulated and fertilized and underwent embryonic death, may contribute to the impairment of fertility observed in these cows. Additionally, it was found that fertility was also impaired by parity independently from polyploidy or aneuploidy, probably as a result of the cow’s aging process.
