Assessing feasibility and cultural characteristics, staining, morphological and biochemical bacterial collection

ABSTRACT

A group of replicas types of bacterial strains preserved at the Bacteriology Laboratory of Research Center of Civil Defense (Enterobacterias: 10, Acinetobacter: 1, Pseudomonas: 1) was studied. The objective was to assess the viability (survival) and original features (cultural, staining, morphological and biochemical features) of a group of microorganisms after 5 years of storage in a sterile mixture of skim milk and glycerol to a final concentration of 10 % and 20 % respectively, contained in vias and cryopreservation at -25 0 °C. All strains studied and their replicas grew in the planting media used to temperature, atmosphere and incubation time required, so 100 % viability was determined. It was found that they maintained their microscopic staining and morphological characteristics, and the same cultural characteristics on solid media and biochemical properties of the moment of conservation. The goods results obtained from the use of this form of conservation provide guaranty and safety to the use of strains in accordance with their functions in biological materials collection; as the continuity of similar studies as a means of testing the major objectives of conservation of microbial cultures, keeping unpolluted alive microorganism in conditions as nearest as possible to the isolation or original characterization.
