Inhibitory activity of alpha-amylase and total phenols in Smallanthus sonchifolius (yacon) ethanolic leaf extracts

ABSTRACT

Introduction: Smallanthus sonchifolius (Poepp. & Endl.) H. Robinson (yacon) is a plant grown in Colombia for its prebiotic and antidiabetic properties.
Objectives: evaluate the alpha-amylase inhibitory activity and the total phenolic content of an ethanolic extract from leaves of yacon grown in Boyacá, Colombia.
Methods: the ethanolic extract was obtained by maceration and percolation of dry pulverized yacon leaves, and was then characterized by phytochemical testing. Total phenolic content was determined by the Folin-Ciocalteu method and results were expressed as mg gallic acid equivalent per dry weight of extract (mg GAE/g DWE) and kg of dry weight of leaves (mg GAE/kg DWL). Alpha-amylase inhibitory activity was determined by the colorimetric method using acarbose as reference inhibitor. Results were expressed as inhibition percentage (I %) and inhibitory concentration 50 (IC50 µg/mL). Data were analyzed with Student's t test.
Results: phytochemical characterization of ethanolic yacon leaf extract points to the presence of phenols, flavonoids, leucoanthocyanidins, tannins, triterpenes, coumarins, lactones, sesquiterpenes and amino acids. Total phenolic content was 7.8 ± 0.4 mg GAE/g DWE and 798.6 ± 45 mg GAE/kg DWL. High inhibitory alphaamylase activity in vitro (CI50= 5.77 ± 0.14 µg/mL) was observed. Results indicate that there is not a significant statistical difference between inhibition percentages for yacon extract and acarbose (p= 0.054).
Conclusion: alpha-amylase inhibitory activity was demonstrated in ethanolic yacon leaf extracts. Such activity could be associated with their high content of phenolic compounds.
