Physicochemical characterization and preliminary diuretic activity of Urera baccifera (L.) watery extracts

ABSTRACT

Introduction: Urera baccifera (L.) Gaudich ex. Wedd (Urticaceae) it is commonly known as chichicate, jamo and ortiga brava. It is an American native wild bush and it has been referred among the plants used in Cuba as diuretic and antilithiasis (the roots), although it has other traditional uses as anti-inflammatory and analgesic. Nevertheless, the scientific studies related with the quality, safety and efficiency of this plant are scarse and those aspects are indispensable to back up its traditional use.
Objective: Evaluate the physicochemical parameters of quality, safety and preliminary diuretic activity of U. baccifera watery extracts of the leaves, stems and roots from Cuba.
Methods: There were obtained watery extracts by decoction and there were determined their quality´s physicochemical parameters. It was analysed the chemical composition by thin layer chromatography, ultraviolet spectroscopy, quantification of phenols and flavonoids by the Folin-Ciocalteu trial and the colorimetric method of aluminum chloride, respectively. It was applied an experimental model in rats to determine the diuretic activity of the extracts to the doses of 400 mg/kg of weight; and metabolic cages to collect urine samples. Finally, it was carried out an oral acute toxicity trial in Wistar rats to evaluate the safety of the extracts.
Results: It was possible to characterize the watery extracts from the physicochemical point of view. In general terms, it was suggested the presence of phenolic compounds finding differences in the total content of phenols and flavonoids. The extracts showed low diuretic effect under the conditions tested that can be compared with the hydrochlorothiazide. The oral acute administration of the watery extracts to the doses of 2000 mg/kg of weight did not produce toxicity in the animals for experimentation.
Conclusions: The study of U. baccifera provided quality, safe and efficient evidences as diuretic and these aspects contribute to justify its use in Cuban traditional medicine. Therefore, it is suggested to carry out broader studies that allow completing these findings.
