Stability of An Anthocyanin-Rich Hydroalcoholic Extract From the Flower of Talipariti elatum (Sw.) Fryxell

ABSTRACT

Introduction: Talipariti elatum (Sw.) Fryxell (majagua) has flowers rich in anthocyanins, a phytochemical used as a raw material in food coloring for human consumption. However, the sensitivity of said pigment to temperatures and pH variations limits its industrial use, making it necessary to study its kinetic stability.
Objective: To evaluate the stability of a hydroalcoholic extract of majagua flowers rich in anthocyanins.
Methods: The flowers were collected and selected manually between October-November 2019, the petals and pistils were separated and frozen at -32 oC. The extract was prepared with mass/solvent 1:2 in 79% v-v hydroalcoholic solution, 0.5% m-v citric acid was added, and it was kept in a sieve for 12 h at 250 revolutions per minute; the mixture was filtered under vacuum, discarding solid residue. pH, total solids, polyphenol and anthocyanin content were quantified in the extract. Stability was evaluated for 21 days at 45 and 55 oC and pH=3 and pH=5, calculating: reaction order (n) of best fit, rate constant (k), half-life time (t1/2), coefficient of temperature (Q10) and activation energy (Ea).
Results: The extract had pH = 3.5; 4.4% total solids; 9.18 mg AGE/ mL (taking the equivalent gallic acid as a standard) of polyphenols and 7.85 mg/L of anthocyanins. Anthocyanin deterioration was fitted to an n=1 model. The greatest stability corresponded to pH=3 and 45 ºC, evidenced at t1/2 =7 days, k=0.09 d-1, Ea=31.7 kJmol-1, and the greatest sensitivity to temperature increase was at pH =3(Q10=1.46). The content of total polyphenols did not change and the degradation of anthocyanins at 55 ºC and pH=5 was greater than 85% after 18 days of storage.
Conclusions: The hydroalcoholic extract of Talipariti elatum was more stable at pH=3 and 45 º C.
