Mechanical stimuli in ovariectomized rats to foster osteogenesis

ABSTRACT

Objective: To determine the number of osteoblasts and observe the bone matrix in rats, subjected to ovariectomy, when applying mechanical stimulation for the control of osteoporosis.
Setting: School of Medicine, Autonomous University of Querétaro, Mexico.
Design: Experimental.
Statistical analysis: Relative risk test with 95% confidence intervals and Chi square with Yate´s correction.
Material and Method: We studied 40 Wistar (Novergicus) rats, average age of 5 months and average weight of 260 g. Rats were subjected to bilateral ovariectomy and divided in two groups: a) experimental group with 20 rats, to which mechanical stimulation was applied, b) control group with 20 rats that did not receive any stimulation. Osteoblasts and bone matrix determinations were performed by means of the Carl Zeiss software (Image analysis).
Results: The bone matrix area of the experimental group was of 6,117 pixels and in the control group of only 3,166 pixels. After application of the mechanical stimulus, 16 rats revealed the presence of osteoblasts, whereas only four rats of the control group (without stimulation) revealed osteoblasts. Relative risk was 4.0 with 95% confidence intervals of 1.62 to 9.87 (p ‹ 0.0005).Proportion of osteogenesis per field in the experimental group was 80% whereas in the control group it was 20%.
Conclusions: Mechanical stimulation in ovariectomized rats favors osteogenesis in type-1 osteoporosis.
